چکیده

The interactions between tubocurarine, pancuronium and alcuronium on neuromuscular transmission have been studied in an isolated neuromuscular preparation (the rat phrenic nerve-hemidiaphragm). Mixtures of tubocurarine and pancuronium, and tubocurarine and alcuronium were found to have a more potent effect than the additive effects of the individual agents when given alone, that is, mixtures were synergistic. Mixtures of alcuronium and pancuronium did not exhibit this phenomenon. Possible mechanisms for these findings are discussed.

Tubocurarine and pancuronium inhibit evoked release of acetylcholine from the mouse hemidiaphragm preparation.

Using a sensitive radioactive method that measures selectively the evoked release of acetylcholine, it was demonstrated that, when stimulating at 50 Hz, tubocurarine or pancuronium 2 X 10(-5) mol litre-1 or hexamethonium 10(-3) mol litre-1 significantly decreased the evoked release of acetylcholine in the mouse in vitro phrenic nerve-hemidiaphragm preparation.

Interactions of diisopropyl phenol (ICI 35868) with suxamethonium, vecuronium and pancuronium in vitro.

In vitro studies were performed using the rat phrenic nerve-hemidiaphragm preparation to investigate possible interactions between diisopropyl phenol and its solvent, cremophor, with three neuromuscular blocking drugs. Cumulative concentration curves were constructed for the neuromuscular blockers and linear regression analyses performed. Differences in the calculated effective concentration to...

Relationships between block-of-twitch and train-of-four fade in the mouse phrenic nerve-diaphragm preparation.

The relationships between the block-of-twitch and train-of-four fade in the presence of nondepolarizing neuromuscular blocking drugs (d-tubocurarine, vecuronium and pancuronium) were examined in vitro by measuring the contractile tension from mouse phrenic nerve-diaphragm preparations. The slope of the block/fade relationship differed between onset of and recovery from neuromuscular block follo...

Mechanisms underlying the prolonged duration of action of muscle relaxants caused by extrahepatic cholestasis.

The neuromuscular blocking effects of Org 6368 and Org NC 45 were studied in rats with experimental cholestasis and in controls. The effect of Org NC 45 during infusion of taurocholate was investigated. In cholestatic rats we observed a three-fold increase of the duration of action of Org 6368 and Org NC 45. The same was observed for Org NC 45 with taurocholate. In the rat phrenic nerve-hemidia...

Age-dependent variation in response to tubocurarine in the isolated rat diaphragm.

The EC50 of tubocurarine was determined in phrenic nerve-hemidiaphragm preparations obtained from 35 Sprague-Dawley rats aged 0-46 days. We measured also the ratio of the fourth to the first twitch in the train-of-four (T4:T1) when the first twitch of the train was depressed to 50% of control. The preparation was not unduly sensitive to tubocurarine at 0 days and there was little evidence of T4...
